IGF-1 LR3 is a 83-amino-acid analog of insulin-like growth factor 1 engineered by (1) extending the native 70-residue sequence with a 13-residue N-terminal peptide and (2) replacing the third amino acid with arginine. These changes reduce its affinity for IGF-binding proteins and protect it from rapid enzymatic cleavage, prolonging plasma half-life from ~10 min to 6โ10 h. Freed from most carrier constraints, IGF-1 LR3 more readily engages IGF-1 receptors (and, to a lesser extent, insulin receptors), triggering the canonical PI3K-Akt-mTOR and MAPK/ERK pathways that drive glucose uptake, protein synthesis, mitosis, and tissue remodeling.
